Rh(D/CE) Genotyping Kit (PCR-SBT method)

Based on the Sanger sequencing typing(SBT), the exon 1-10 allele sequences of RhD and RhCE in DNA extracted from human whole blood or tissue were detected. Based on the interpretation results of the International Society for Blood Transfusion (ISBT) database, RhD and RhCE blood groups were predicted, providing reference for RhD and RhCE gene research.
The Rh blood group system is composed of 56 antigens. The RhD gene and RhCE gene encoding RhDCcEe blood group antigen are located in the 34.3-36.1 region of the short arm of human chromosome 1, closely connected and highly homologous (93.8%). They both have 10 exons and 9 introns, with lengths of 57932 bp and 58575 bp, respectively, encoding 417 amino acids. The exon 8 is completely identical, with significant differences in exon 3, 4, 5, 7, 9, and intron 4. Complete deletion of exons, mutual substitution, base mutations lead to weakened or missing related antigens. RhD, C, c, E, and e antigens all have strong immunogenicity and can cause varying degrees of hemolytic transfusion reactions and neonatal hemolysis
Sanger sequencing typing(SBT) uses human genome DNA as a template and uses specific primers to amplify corresponding target fragments. Perform electrophoretic testing on amplified products to verify the effectiveness of amplification(this step is quality control process). The purified amplification product is used as template for sequencing amplification. Sequencing reactions use both forward and reverse specific primers. By comparing the sequence results with the publicly available human RhD and RhCE standard gene sequence information in the International Society for Blood Transfusion (ISBT) database, genotyping results were obtained. The allele names follow the naming principles of the ISBT Red Cell Immunogenetics and Blood Group Nomenclature Committee (ISBT 004), RhD (v6.2 30-SEP-2022), and RhCE (v6.2-31-MAR-2022). Primers were designed according to the sequence information published in Genbank database of NCBI.
