Exosome-Human CD81 Isolation Reagent (from Cell Culture) enables the isolation of CD81-positive cells from enriched cell culture media Extracellular vesicles (ECVs) such as exosomes. Once isolated, specific subpopulations of exosomes/ECVs can be further studied using methods such as Western blotting, qPCR, or next-generation sequencing.
• Minimize hands-on time for specific exosome isolation
• Obtain highly pure exosome subsets
• Protocol is easily scalable
• Compatible with most downstream analyses
Recent studies have shown that the exosome/ECV population consists of several subsets that may have different functions. These subsets can be selectively isolated and studied using immunoaffinity-based Dynabeads™ magnetic separation technology. The cell culture medium must first be enriched for exosomes. This can be done quickly and efficiently using
Total Exosome Isolation Reagent (from cell culture media) or traditional ultracentrifugation . After enrichment, specific exosomes are isolated by binding to the surface of magnetic beads.
Dynabeads™ magnetic beads are known for their sensitivity, reproducibility and stability. Dynabeads™ magnetic separation technology also helps you “see” your sample during the process because of the color of the beads. When the sample tube is placed in the magnetic stand, the bead-bound exosomes are pulled to the sides of the sample tube, allowing for easy separation and buffer exchange. Additionally, sample and bead volumes can be easily expanded based on your sample size or downstream application.
Note: Be sure to mix thoroughly to ensure the magnetic beads do not settle in the sample tube. We recommend a mixer that offers tilt and rotation for optimal mixing.
