Use this kit to enrich viable mouse dendritic cells (DC) from spleen/lymph node cells by negative isolation. This kit removes T cells, mIgM+ B cells, NK cells, red blood cells, and most granulocytes. Highly enriched mouse DCs are bead- and antibody-free and can be used in any functional assay. Cells can be further sorted to obtain highly pure specific DC subpopulations.

Recovery of viable Lin-CD11c+ DC cells was high (80-100%) across all subpopulations, including DCs positive for CD4, CD8α, B220, and CD19.

Method:
Add the anti-non-dendritic cell antibody cocktail to the sample and allow to bind to the cells. Dynabeads™ are added and bind to antibody-labeled cells during a short incubation period. Bead-bound cells on the magnetic rack are rapidly detached and discarded. The remaining unexposed mouse DCs can be directly sorted into selected subpopulations or used for any other application.
