PureLink™ Viral RNA/DNA Miniprep Kit is a nucleic acid purification system designed for purification of cell-free samples such as Quickly and easily isolate viral RNA or DNA from serum, plasma, and cerebrospinal fluid). The starting sample volume can be as high as 500 µL, and the elution volume can be as low as 10 µL, achieving 50-fold concentration of nucleic acids. The higher target concentrations achieved using the PureLink™ Viral RNA/DNA Miniprep Kit provide greater sensitivity and accuracy in downstream detection procedures such as real-time PCR or endpoint analysis. Specially formulated buffers enable the purification of RNA or DNA, allowing one system to be used for all virus extraction samples. Additionally, the kit provides sufficient lysis buffer for larger starting sample volumes.

Samples prepared using the PureLink™ Viral RNA/DNA Miniprep Kit have consistently lower Ct values (Figure 2) , demonstrating higher sensitivity
Furthermore, quantitation was accurate down to 20 pfu compared to samples prepared using the supplier's Q kit (stabilized at 200 pfu). Results showed improved viral RNA recovery and greater overall consistency in quantitative amplification.
Ct values obtained for DNA prepared using the PureLink™ Viral RNA/DNA Miniprep Kit were lower compared to results obtained using the supplier's Q kit (Figure 3), indicating sensitivity in detecting viral DNA higher. The data shows that the PureLink™ Viral RNA/DNA Miniprep Kit is able to recover DNA down to 20 pfu with a significantly lower Ct value compared to competing kits.
The PureLink™ Viral RNA/DNA Miniprep Kit produces target molecules with consistently lower Ct values compared to the EZ1 Kit; in some cases, up to 4 cycles earlier (Figure 4). Arrows indicate samples that failed to produce Ct values. A total of 14/48 samples failed to produce Ct when processed on the BioRobot EZ1 (29%), while the PureLink™ Viral RNA/DNA Kit failed to provide Ct values in only 3 samples (6%) using Two replicate samples extracted with the EZ1 kit also failed to produce Ct values.
For enhanced detection of viral RNA or DNA, the PureLink™ Viral RNA/DNA Miniprep Kit has been shown to provide lower Ct values and higher Ct values compared to competing products using supplier-recommended volumes. PureLink™ Viral RNA/DNA Miniprep Kit contains micro spin column in collection tube, collection tube, recovery tube, lyophilized carrier RNA , Proteinase K, Lysis Buffer, 5X Wash Buffer II, and RNase-free water. Each kit contains enough material for 50 purifications. Kits ship at room temperature; upon receipt, store lyophilized vector RNA at -20°C and proteinase K at 4°C. When stored as directed, the kit is stable for 1 year.
