DMEM (Dulbecco's Modified Eagle Medium) is a widely used basal medium that can be used to support many types of mammalian cells grow. Using DMEMDMEM differs from other media in that it contains 4 times the concentration of amino acids and vitamins equivalent to the original Eagle Minimum Essential Medium. DMEM is initially formulated with low concentrations of glucose (1 g/L) and sodium pyruvate, but is often used in high glucose environments with/without sodium pyruvate. DMEM contains no proteins, lipids or growth factors. Therefore, DMEM needs to be supplemented with nutrients, usually 10%
Fetal bovine serum (FBS). DMEM uses a sodium bicarbonate buffer system (3.7 g/L) and therefore requires a 5–10% CO
2 environment to maintain physiological pH. Gibco™ cell culture media in powder form requires supplementing sodium bicarbonate, adjusting pH, and filtering (see
