Thermo Scientific QuantaRed Enhanced Chemifluorescent HRP Substrate (ADHP) is the most sensitive fluorescent ELISA substrate available and provides enhanced Type chemiluminescence with considerable sensitivity. The kit contains QuantaRed ADHP Concentrate, QuantaRed Enhancer Solution, QuantaRed Stabilized Peroxide Solution, and QuantRed Stop Solution and is sufficient for 960 wells (10 x 96-well plate, 100 µL working solution/well).
Features of QuantaRed Enhanced Chemifluorescence HRP Substrate Kit:
•Sensitive — low picogram to femtogram level detection
• Redshift - Minimal interference from autofluorescence from biological samples
• Convenience — Compatible with existing filter sets, no need to purchase special filter sets
• Supplied as a complete set — Mix and use reagents, no need to prepare buffers or dissolve reagents
• Flexible — Use stop solution to effectively terminate reactions for at least 4 hours or more Stable signal over long periods of time.
•Universal — Generates a strong colorimetric signal, reading fluorescence or colorimetric signal at 570 nm
QuantaRed HRP substrate can be used in ELISA specifications for peroxidase Efficient and sensitive fluorescence and colorimetric detection of activity. QuantaRed substrates use patented technology to improve fluorescence yield and sensitivity of the well-documented ADHP (10-acetyl-3,7-dihydroxyphenoxazine) chemofluorescence reaction. ADHP is a nonfluorescent compound that reacts with horseradish peroxidase (HRP) to produce resorufin, a soluble highly fluorescent reaction product (maximum excitation/emission wavelength ∼570/585 nm). The long wavelength emission of resorufin minimizes interference from lower wavelength (blue and green) autofluorescence, making it an ideal substrate for the quantitative determination of target molecules in biological samples. Furthermore, the reaction product of QuantaRed substrate is colorimetric and fluorescent and can be read spectrophotometrically at ∼570 nm.
Specially formulated QuantaRed substrate contains chemical enhancers for maximum sensitivity and excellent low-end linearity. QuantaRed enhanced HRP substrate has higher quantum yield and larger extinction coefficient, and its sensitivity is comparable to enhanced chemiluminescence. It is more sensitive than Amplex red substrate when detecting HRP in ELISA applications.
