Thermo Scientific NHS-PEG4-Azide is an amine-reactive compound that can be used to derivatize proteins with primary or amines Coated polymer surface for attachment to phosphine-modified molecules.
The NHS ester compounds can react with primary amines (eg, lysine residue side chains or aminosilane-coated surfaces) to form covalent bonds. The azide (N3) group reacts with phosphine-labeled molecules through a mechanism called Staudinger chemistry, enabling efficient and specific coupling of derivatized molecules in biological samples.
Characteristics of NHS-PEG4-azide:
•
Solubility—The reagent is readily soluble in water-soluble solvents and is used in combination with cell lysates and other organisms Subsequent dilution of sample in aqueous reaction mixture
•
Compatibility—Efficient chemical reaction under simple buffer conditions; no auxiliary reagents such as copper or reducing agents are required
• < b>Specificity—NHS ester is designed to covalently bind to a primary amine (the side chain of a lysine or the N-terminus of a polypeptide)
•
Chemiselectivity—Azide Compounds and phosphine groups do not react with or interfere with components of biological samples (bioorthogonality), but efficiently couple to each other
