TMB (3,3',5,5' tetramethylbenzidine) is a chromogen that produces blue when oxidized by hydrogen peroxide (catalyzed by HRP), with maximum absorption wavelengths at 370 and 652 nm. After adding sulfuric acid or phosphoric acid to stop the reaction, the color changes to yellow (measured at 450 nm). TMB is very sensitive and is oxidized faster than other HRP substrates, resulting in faster color development. Unlike other commercially available substrates, these products do not contain DMF or DMSO. There are three formulas, the main difference being sensitivity. Slow TMB has medium sensitivity and is suitable for kinetic readings. The sensitivity of Turbo TMB is comparable to that of approximately 1 mg/mL OPD. Ultra TMB-ELISA produces extremely high signal-to-noise ratio and sensitivity in the picogram range.
