Thermo Scientific Pierce Metal Enhanced DAB Substrate Kit in a specially formulated diaminobenzidine peroxidase substrate Cobalt chloride and nickel chloride are used to produce darker colors in immunohistochemical staining.

To prepare the product, use Pierce Stabilized Peroxide Buffer before use The 10X metal-enhanced substrate solution is diluted with liquid. The special formula makes the 1X substrate stable at room temperature for at least 6 hours. There is no increase in background color of the substrate, and unlike typical metal-enhanced DAB methods, reactivity remains unchanged (1,2). The resulting precipitate is a dark brown-black color, darker than that produced by DAB alone. Not only does the intensity increase, but the background value also decreases, which increases the signal-to-noise ratio and makes screening tissue or membrane antigens easier.
