Cell Proliferation Dye eFluor™ 670 is a red fluorescent dye that can be used to monitor single cell division. This fluorescent dye binds to any cellular protein that contains a primary amine, and as cells divide, the dye is evenly distributed among daughter cells, as measured by the sequential halving of the dye's fluorescence intensity. Up to 6 passages of cells were observed. Cell Proliferation Dye eFluor™ 670 can also be used for long-term tracking of labeled cells. Using two-parameter plot analysis provides better discrimination for each generation of cells, especially between undivided cells and primary cells. Cells labeled with Cell Proliferation Dye eFluor™ 670 are fixed and permeabilized for intracellular target analysis using standard formaldehyde-containing fixatives and saponin-based permeabilization buffers such as the Foxp3 Transcription Factor Staining Buffer Kit (Cat. No. 00-5523) or IC Fixation Buffer (Cat. No. 00-8222) and Permeabilization Buffer (10X) (Cat. No. 00-8333).

Cell Proliferation Dye eFluor™ 670 has an excitation peak of 647 nm and can be excited by the red (633 nm) laser line. It has an emission peak at 670 nm and can be detected using a 660/20 bandpass filter (equivalent to APC, Alexa Fluor™ 647 or eFluor™ 660), making it compatible with applications utilizing GFP.

Cell Proliferation Dye eFluor™ 670 has a molecular weight of 792.6 and is supplied as a lyophilized powder. The lyophilized powder can be reconstituted with 126 µL of anhydrous DMSO per vial to a 5 mM stock solution; after reconstitution, it should be stored with desiccant at -20°C in the dark. It is recommended to use the reconstituted dye within 6 months and avoid repeated freezing and thawing.
