JC-1 is a membrane-permeable dye widely used in flow cytometric analysis and fluorescence microscopy. Determine mitochondrial membrane potential. This dye selectively enters mitochondria, where it changes color reversibly as membrane potential increases (potential values above approximately 80-100 mV). This property arises from the reversible formation of JC-1 aggregates upon film polarization, which when excited at a wavelength of 488 nm causes the emission to shift from 530 nm, the emission wavelength of the monomeric form of JC-1. Move to 590 nm (the emission wavelength of J-polymer). Both colors can be detected using separate filters for FITC and PE/Phycoerythrin/Rhodamine. JC-1 can be used for both qualitative (an observable shift in fluorescence emission from green to orange) and quantitative (fluorescence intensity can be measured on two filter sets) analyses. JC-1 can be used to indicate whether apoptosis has begun.
