Ideal for immunoprecipitation of protein complexes:
Low background and covalent attachment of antibodies to the bead surface make Dynabeads Tosylactivated ideal for immunoprecipitation of proteins and protein complexes ( Excellent choice for co-immunoprecipitation, co-IP). Gentle and rapid magnetic aggregation of beads and extremely short incubation (possibly due to fast surface binding kinetics) make Dynabeads Tosylactivated ideal for immunoprecipitation of extremely unstable or and/or transient (ephemeral) protein complexes.

Dynabeads Tosylactivated Microbeads Surface Properties:

• p-Tosyl-sulfonyl (Tosyl) group
• Hydrophobic, pH neutral
• Covalent binding via primary amine (NH2) or sulfhydryl (SH) groups

Overview of coupling process:
By overnight incubation Optional ligands and Dynabeads Tosylactivated achieve covalent coupling. Ligands coupled to Dynabeads Tosylactivated typically include peptides and proteins (such as antibodies used for immunoprecipitation or co-immunoprecipitation). Coupling occurs at neutral to high pH and 37°C. We recommend coupling at pH 8.5-9.5, but for ligands with poor pH stability, coupling can be performed in other buffers at pH 7.4.

Once the ligand coupling step is complete, the actual surface coating of Dynabeads Tosylactivated is no longer active and therefore non-specific binding is low.

Dynabeads are nonporous monodisperse superparamagnetic microbeads. They are highly mobile in solution, allowing ligands coupled to the beads to continuously interact with the entire sample. Transfer the test tube to a tube rack containing a strong magnetic field, and the superparamagnetic beads will adsorb to the tube wall. The strong magnetic field quickly adsorbs the beads to the tube wall, so all the supernatant can be easily removed by pipetting. The washing steps are similar.
