Thermo Scientific™ Pierce™ Protein A/G Magnetic Sepharose Beads provide a fast and convenient method for isolation from serum, cells Immunoglobulins were purified from culture supernatant or ascitic fluid. For antibody purification, magnetic beads are incubated with the antibody sample and then magnetically separated from the supernatant. Nonspecifically bound serum or host proteins can be washed away, and bound antibodies can be separated using an elution buffer. Remove magnetic beads from solution using a magnetic stand in manual applications or an instrument such as the Thermo Scientific™ KingFisher™ Flex Magnetic Bead Purifier in automated applications. Automated instruments are particularly useful for higher throughput purification and screening purification conditions.

Pierce Protein A/G Agarose Magnetic Beads contain a A 50.5 kDa protein A/G recombinant fusion protein covalently linked to a magnet-embedded agarose core particle. These magnetic beads are not simply a mixture of separate protein A and protein G peptides and then immobilized, nor are they a mixture of protein A magnetic beads and protein G magnetic beads. Recombinant chimeric protein A/G combines four IgG binding domains of protein A and two binding domains of protein G, making it a more versatile and convenient tool for purifying immunoglobulins.

Features of Protein A/G Sepharose Magnetic Beads:
•Protein A/G – Immobilized recombinant fusion proteins of the antibody binding domains of Protein A and Protein G can be obtained from almost any mammalian species Purified IgG
• High capacity – sufficient for routine and demanding purification procedures
• Low non-specific binding – optimized purification protocols for better IgG purification
• Flexibility – simultaneously on one magnetic bead Contains IgG binding domains for Protein A and Protein G for convenience
• Compatibility—Magnetic beads are compatible with manual and automated applications
• Inert and stable—Excellent production method immobilizes proteins via leach-resistant covalent bonds A/G

High performance, magnetized superparamagnetic agarose beads are validated and optimized for use on high-throughput magnetic platforms such as the KingFisher 96 and KingFisher Flex Magnetic Bead Purifiers, however, magnetic beads Excellent performance can also be achieved for simple benchtop purification applications with the appropriate magnetic stand.

Our recombinant Protein A/G binds to all human IgG subclasses, making it ideal for purifying monoclonal or polyclonal IgG antibodies when subclass characterization is undefined. In addition, it binds IgA, IgE, IgM and IgD (the latter is weakly binding). Protein A/G binds adequately to all mouse IgG isoforms but does not bind mouse IgA, IgM or serum albumin. Therefore, Protein A/G is an excellent choice for purifying and detecting monoclonal antibodies of mouse IgG subtypes, avoiding the interference of IgA, IgM and mouse serum albumin. Some subclasses of mouse monoclonal antibodies have higher affinity to recombinant protein A/G than protein A or protein G.
