Thermo Scientific Halt Protease Inhibitor Cocktail (EDTA-Free) (100X) is a ready-to-use protease inhibitor concentrated stock solution , can be added to samples to prevent proteolytic degradation during cell lysis and protein extraction.
Halt Protease Inhibitor Cocktail (EDTA-Free) (100X) Features:
•
Convenience—Freezer-stable 100X liquid strength is greater than either inhibitor alone or Other strengths are more effective and easier to use; just measure exactly what you need
•
No Proprietary Ingredients—Fully disclosed formula contains optimized concentrations of six broad-spectrum protease inhibitors Agents AEBSF, aprotinin, statin, E-64, leupeptin and gastropin A, stable in high quality dimethyl sulfoxide (DMSO)
•
Multiple package sizes b>—Available in a variety of package sizes, including single-use microtubes that keep protease inhibitors fresh and reduce the risk of reagent contamination
•EDTA-free formulation—Ensures compatibility with isoelectric focusing
br>•Lysis Buffer Compatible—Use with Thermo Scientific Pierce Cell Lysis Buffer or virtually any other commercial or homemade detergent-based lysis reagent. Add 10 µL of concentrated mix/1 mL of lysis buffer to ensure complete protection of the resulting protein extract.
Applications:
•Protection of intact active cellular proteins from degradation by endogenous proteases
•Inhibition of protease activity of lysates produced by a variety of methods, Includes Thermo Scientific Pierce Cell Lysis Reagent, other commercially prepared cell or tissue lysates, sonication, French press, mixer homogenization, or repeated freeze-thaw cycles
• Screening of extract proteolytic activity
• Studying Proteolysis in the Regulation of Cellular Processes
Proteases constitute a large class of enzymes, including endo- and exopeptidases that catalyze the hydrolytic breakdown of proteins into peptides or amino acids in a variety of cell and tissue types Enzymes. Various proteases are regulated and compartmentalized in living cells, but cell lysis releases them and mixes them with other extracted proteins. Therefore, their activity can lead to the loss of large amounts of valuable proteins, negatively affecting downstream applications. Therefore, it is common practice to puncture cell lysis buffers or cell extracts with known mixtures of compounds to inactivate or inhibit various proteases. Halt Protease Inhibitor Cocktail effectively inhibits serine, cysteine, aspartic and aminopeptidase proteases commonly found in cell lysate samples. Regular and EDTA-free formulations inhibit various metalloproteases and are compatible with 2D electrophoresis, respectively.
