Because the antibodies are covalently attached to the beads, IP targets can be easily eluted and recovered without antibody contamination. The c-Myc peptide (EQKLISEEDL) derived from the C-terminal region of the human c-Myc protein is one of several fusion protein tags used for recombinant protein expression. Pierce c-Myc Magnetic IP/Co-IP Kit uses specific, high-affinity immobilized antibodies (clone 9E10) for bacterial and mammalian cell lysates and using Pierce HumanIn VitroTranslation Reagents Rapid immunoprecipitation of c-Myc-tagged fusion proteins from box-prepared lysates. The beads are incubated with cell lysate containing c-Myc-tagged protein to capture the fusion protein, and the beads are subsequently washed and eluted using either a low pH elution buffer or a non-reducing sample buffer. Protocols and buffers are optimized to maximize the efficiency of IP and co-IP reactions and to enrich specific protein-interacting complexes in eluted samples. Anti-c-Myc antibodies can be used to detect c-Myc-tagged proteins by Western blot analysis.
