Gibco FreeStyle F17 Expression Medium is a chemically defined, protein-free, serum-free medium developed to support Growth and transfection of HEK293 cells in suspension culture. Requires 4-8 mM L-Glutamine or GlutaMAX supplement before use.

Little adaptation required
Suspension grown 293-F, 293-H , FreeStyle 293-F cells can be cultured directly in FreeStyle F17 expression medium without adaptation. Cells adapted to other commercially available serum-free media can be subcultured directly in FreeStyle F17 media, usually without any further adaptation. Cells often need to adapt to serum-containing formulas.

Growth and transfection of HEK293 cells
FreeStyle F17 Expression Medium supports 293-F cells at a peak density of 6-7 x 10⁶ viable cells/mL. Cells cultured in FreeStyle F17 medium can be transfected using a variety of transfection reagents, including lipofection reagents such as 293fectin and FreeStyle MAX reagent.

Scalable in spinner bottles and bioreactors
Production using FreeStyle F17 Expression Medium can be scaled up in spinner bottles or bioreactors. The appropriate rotator or impeller speed and inoculum density should be optimized for each system. Depending on impeller design and rotational speed, it may be necessary to supplement FreeStyle F17 medium with additional Pluronic F-68 to avoid shear stress in the culture.
