Thermo Scientific EquiPhi29 DNA Polymerase is a proprietary phi29 DNA polymerase mutant created through in vitro protein evolution. With phi29 DNA polymerase, the thermal stability, reaction speed, product yield, and amplification bias of this enzyme's protein have been significantly improved, while retaining all the advantages of the wild-type enzyme, including high processivity (greater than 70 kb), strong strand displacement activity and 3'→5' exonuclease (proof reading) activity that preferentially acts on single-stranded DNA or RNA. Therefore, the use of hydrolysis-resistant random primers is recommended.
Product advantages include:
• Lowest amplification bias on the market
• Higher yields of amplified DNA, even using micro-volume templates
• In less time Enable highly accurate DNA synthesis
Applications:
• Unbiased whole genome amplification (WGA) using multiple sample types:
– DNA from single cells
– Uncultured Microbial cells and virus particles
– Pathogenic microorganisms or metagenomes
– DNA for SNP and STR detection
• Rolling circle amplification (RCA)
• Protein primer DNA amplification
• Cell-free cloning of lethal DNA
• In situ genotyping with padlock probe
• RNA-primed DNA amplification
Note: Add pyrophosphate to the reaction mixture containing EquiPhi29 DNA polymerase Enzymes may further enhance DNA synthesis.
