FirstChoice™ RLM-RACE Kit is designed to amplify cDNA from only full-length, capped mRNA, typically after PCR Generate a single band. This kit is a major improvement on the basic Rapid Amplification of cDNA Ends (RACE) protocol. The RLM-RACE program selects only full-length mRNA (not rRNA, tRNA, or degraded RNA) and facilitates the cloning of sequences from the 5' end of the message.

RACE
Rapid amplification of cDNA ends (5'-RACE) is a polymerase chain reaction-based technology that facilitates the cloning of sequences from the 5' end of the message. The FirstChoice™ RLM-RACE Kit is a major improvement over the basic RACE protocol.

Select full-length mRNA (not rRNA, tRNA, or degraded RNA)
In the FirstChoice™ RLM-RACE procedure, by using calf intestinal phosphatase (CIP) Total RNA or poly(A) RNA removes 5'-phosphate from all molecules containing free 5'-phosphate (ribosomal RNA, fragmented mRNA, tRNA, and contaminating genomic DNA) to select for full-length mRNA. Full-length mRNA is unaffected. The RNA is then treated with tobacco acid pyrophosphatase (TAP) to remove the cap structure in the full-length mRNA, leaving the 5'-monophosphate. A synthetic RNA adapter is ligated to the RNA population (molecules containing only 5'-phosphate), and the uncapped full-length mRNA will accept the adapter. Then, random primer labeling, reverse transcription reaction, and nested PCR were performed to amplify the 5' end of specific transcripts.

SuperTaq™ Thermostable Taq DNA Polymerase is available separately. For optimal amplification of ≥1 kb fragments, use SuperTaq-Plus. Also included are 3' RACE adapters and primers for 3' RACE.
