Ambion™ NorthernMax™-Gly Kit is an alternative to formaldehyde-based northern blot analysis using glyoxal/ A loading solution of DMSO was used for sample denaturation. The reagents provided are sufficient to treat 1000 cm² membranes.

Using glyoxal/DMSO instead of formaldehyde to prepare samples avoids pouring and running gels in a fume hood and eliminates safety concerns associated with the use of formaldehyde. Likewise, RNA samples denatured with glyoxal may show clearer bands on a Northern blot than denatured samples, and can subsequently be electrophoresed in the presence of formaldehyde. Simplified NorthernMax™-Gly procedure is more convenient and faster than standard glyoxal gel protocols. A single reagent is used for sample denaturation and loading, which shortens the incubation time for sample denaturation and eliminates the need for buffer recirculation during electrophoresis. The volume ratio of glyoxal denaturing solution to sample RNA is lower than other published protocols, so sample precipitation is usually not required before gel loading. NorthernMax™-Gly Kits can be used with radiolabeled or non-isotopically labeled RNA or DNA probes. Each NorthernMax™-Gly kit provides a complete system. You must purchase the positively charged nylon membrane separately (see accessories). RNA samples and templates for probe synthesis are also provided as controls for each kit.
