Thermo Scientific T4 RNA Ligase catalyzes the intermolecular and intramolecular 5'-phosphate transfer of oligonucleotides, single-stranded RNA and DNA ATP-dependent formation of a phosphodiester bond between the radical and the 3'-hydroxyl terminus.
Minimum substrate: a nucleoside 3',5'-hydrogen phosphate in intermolecular reactions and an 8-base oligonucleotide in intramolecular reactions.
Applications:
•RNA 3'-end labeling (using cytidine 3',5'-bis[α-32 P]phosphate)
• RNA-to-RNA ligation
• Synthesis of oligoribonucleotides and oligodeoxyribonucleotides
• tRNA-specific modification
• Oligodeoxynucleotide ligation to single stranded cDNA to achieve 5' RACE (Rapid Amplification of cDNA Ends)
• Site-specific generation of PCR complex primers
