Thermo Scientific S1 Nuclease degrades single-stranded nucleic acids, releasing 5'-phosphoryl mononucleotides or oligonucleotides . It is five times more active on DNA than on RNA. S1 nuclease can also cleave dsDNA at single-stranded regions caused by gaps, clefts, mismatches, or loops. S1 nuclease exhibits 3'-phosphomonoesterase activity.
The enzyme is a glycoprotein with a carbohydrate content of 18%.
Applications:
• Removal of single-stranded overhangs from DNA fragments
• S1 transcript localization
• Hairpin loop cleavage
• Works with Exonuclease III to create unidirectional deletions in DNA fragments

Under conditions of high enzyme and low salt concentrations, S1 nuclease can introduce breaks Double-stranded DNA, RNA and DNA/RNA hybrids.
