Thermo Scientific Endonuclease IV (Endo IV) recognizes apurine/apyrimidine (AP) sites in dsDNA and cleaves binding to Damage the phosphodiester bond 5' to the site, creating a hydroxyl group at the 3'-terminus (SeeFigure 1 in the Supporting Data). This enzyme also acts as a 3'-diesterase, capable of releasing 3'-phosphoglycolic acid or 3'-phosphate from damaged ends of dsDNA. Endo IV also has 3'→5' exonuclease activity. The synthesis of this enzyme on the substrate is very sensitive to ionic strength, metal ions, EDTA and reducing conditions. Substrates with 3'-recessed ends are preferred substrates for 3'→5' exonuclease activity.
This enzyme does not require Mg2+ and is fully active in the presence of moderate concentrations of EDTA.
