Thermo Scientific WELQut Protease is a highly specific recombinant serine protease from Staphylococcus aureus.
It recognizes and precisely cleaves recombinant proteins containing engineered recognition sequences † W- E- L- Q↓X (Trp, Glu, Leu, Gln, X can be any amino acid). The protease cleaves outside the recognition sequence, leaving no additional amino acids bound to the target protein.
WELQut protease is active over a wide range of temperatures (4 to 30°C) and pH (pH 6.5 to 9.0) and does not require specific buffers.
In addition, this new protease offers several procedural advantages - it is suitable for on-column proteolysis reactions and can be easily removed from the reaction mixture using a built-in His-tag.
Product Advantages•Cleaving outside the WELQ recognition sequence will not cause additional amino acids to bind to the target protein
•For homologous recognition sites Highly specific and does not generate non-specific product bands even after long incubations and excess protease use
• Can be easily removed from the reaction mixture using the built-in His- tag
• Suitable for columns Proteolytic Reaction
Applications•Remove N-terminal fusion tags from recombinant protein preparations.
Note†This cleavage sequence is present in the expression vector pLATE52, included in
Thermo Scientific aLICator LIC Cloning and Expression Kit 4 (N-Terminal His Tag/WELQut) available from Thermo Scientific (#K1281).
