Thermo Scientific terminal deoxynucleotidyl transferase (TdT) is a template-independent DNA polymerase that catalyzes deoxyribose sugar Nucleotide repeats are added to the 3'-OH of oligodeoxyribonucleotides and single- and double-stranded DNA. TdT requires an oligonucleotide consisting of at least three nucleotides as a primer. When using RNA as a template, TdT shows different properties, mainly depending on the tertiary structure of the 3'-end of the acceptor RNA and the nature of the nucleotide. Typically, performance is lower than when using DNA as a template.

Remarks
TdT reaction buffer is not compatible with downstream applications due to the presence of CoCl2. CoCl2 from the reaction mixture must be removed by spin column or phenol/chloroform extraction and subsequent ethanol precipitation.
