Note: Homing endonuclease does not have a strictly defined recognition sequence. They tolerate minor sequence changes that only partially affect the cleavage reaction. I-SceI may bind to severed DNA. This can cause DNA band drift during electrophoresis. To avoid atypical DNA banding patterns, use 6X DNA loading dye with SDS solution for sample preparation, or heat digested DNA in the presence of SDS before electrophoresis. Enzyme activity detection uses pUC-I-SceI DNA.
