Thermo Scientific SfiI restriction enzyme recognizes the GGCCNNNN^NGGCC site and cleaves best in G buffer at 50°C. Note: FastDigest enzyme is also available for fast DNA digestion.

Thermo Scientific Conventional Restriction Endonucleases are a large collection of high quality restriction enzymes optimized to function in one buffer of a five-buffer system. In addition, a universal Tango buffer is provided to facilitate double enzyme digestion. All enzymes demonstrated 100% activity under recommended buffers and reaction conditions. To ensure consistent performance, Thermo Scientific Restriction Enzyme Reaction Buffers contain premixed BSA, which enhances the stability of multiple enzymes and binds contaminants that may be present in DNA preparations.

Note: The enzyme activity test uses pUC19 DNA with an inserted fragment, which contains two SfiI sites derived from adenovirus-2 DNA point. 10% active when incubated at 37°C. Overlapping dcm methylation results in reduced SfiI digestion. To avoid dcm methylation, use a dam-, dcm strain such as GM2163. When using SfiI for digestion, at least two copies of its recognition sequence are required. The two sites can be on the same or different DNA molecules. (LM Wertzell et al., J. Mol. Biol, 248, 581-595, 1995.) Therefore, an oligonucleotide containing a SfiI recognition site can also be added. See product specifications for methylation sensitivity.
