Thermo Scientific Phusion Plus DNA Polymerase is a hot-start, high-fidelity DNA polymerase that uses protein fusion technology and has universal Primer annealing function. It is a Pyrococcus -like proofreading DNA polymerase fused to a processivity-enhancing domain, allowing Phusion Plus DNA Polymerase to generate PCR sequences with high accuracy, sensitivity, and inhibitor tolerance.

New Phusion Plus reaction buffer contains proprietary additive that stabilizes the primer/template duplex during the 60°C annealing step. This unique buffer formulation improves PCR specificity by eliminating the need to optimize annealing temperatures for each primer pair individually, helping to save time and avoid human error in PCR runs. Additionally, with Phusion Plus DNA Polymerase and its universal annealing capabilities, different PCR reactions can be cycled simultaneously using the same protocol.

Phusion Plus DNA Polymerase is inactivated at room temperature due to a parent molecule-mediated hot-start mechanism, allowing for preparation and storage of PCR reactions prepared at room temperature. Enzyme activity is not fully activated until the predenaturation step.
