Simple purification of RNA from whole blood PureLink™ Blood Total RNA Kit is designed to purify from 50 to 500 µL of fresh whole blood, or approximately 3.5 x 10
6 leukocytes (leukocytes), in less than 1 hour. The entire protocol can be completed in centrifuge tubes and spin columns.
Leukocyte enrichment for improved RNA purification Whole blood consists of anucleate red blood cells (erythrocytes, 4 - 6 million/mm
3) and white blood cells, with the latter accounting for a minority (5000 - 7000/mm
3). Purification of total RNA from whole blood without removal of red blood cells will result in lower RNA yields and clogging of the purification column. A unique feature of the PureLink™ Blood Total RNA Kit is its use of a leukocyte enrichment step. Hypotonic shock is used to selectively lyse red blood cells without affecting white blood cells. Anucleate red blood cells are efficiently removed, significantly reducing cell debris that could interfere with RNA purification and other downstream applications. Therefore, high-quality total RNA can be obtained using the PureLink™ Blood Total RNA Kit.
