PureLink™ HiPure Plasmid Filtration Maxiprep Kit is designed to isolate higher purity from 100-500 mL overnight bacterial cultures Plasmid DNA. PureLink™ HiPure Plasmid Filtration Kits include HiPure filtration columns that combine purified bacterial lysate filtration and plasmid DNA anion exchange resin purification in a single device. The quality of the resulting plasmid DNA was comparable to that obtained by two cesium chloride gradient purifications, a more stringent method of DNA purification. For transfection and other applications, purify high-quality DNA in less than 2 hours without additional steps to remove contaminants such as RNA, proteins, or endotoxins. In addition, phenol, chloroform, ethidium bromide and cesium chloride are removed from the protocol, minimizing exposure and handling of hazardous materials. How it works
Add HiPure filter columns to PureLink™ HiPure Plasmid Kits , which quickly clarifies bacterial lysates without the need for centrifugation (see figure). The lysate filter is integrated into the DNA binding column to combine the steps of lysate purification and direct binding of plasmid DNA to the anion exchange resin. A single column wash removes impurities such as RNA, proteins, metabolites, and other low molecular weight molecules; ultrapure plasmid DNA is then eluted with high-salt buffer. The DNA was then desalted and concentrated by an isopropanol precipitation step and then collected by centrifugation. The entire program can be completed in approximately two hours.
