ProLong Gold Antifade Mounting Medium is a mounting medium that can be used directly on fluorescently labeled cells or tissue samples on microscope slides. Liquid mounting medium. Its formula does not contain DNA stains or contains the blue DNA stain DAPI or the deep red DNA stain SYTOX Deep Red. ProLong Gold Mounting Medium contains chemicals that protect fluorescent dyes from quenching (photobleaching) during fluorescence microscopy experiments, providing this protection without significant quenching of the initial fluorescent signal. It is a curing mounting medium that creates a near-perfect optical pathlength (1.47 RI), ensuring long-term storage of samples. It's ready-to-use (store at room temperature and requires no mixing)—just drop in sample, cover with a slide, and it's ready to cure and image.

It is not recommended to use ProLong Gold anti-fade mounting medium to mount samples containing fluorescent proteins (such as GFP). To provide excellent antifade protection for fluorescent proteins and fluorescent dyes, ProLong Diamond Antifade Mounting Medium is recommended. Learn more about the key differences between ProLong Gold Antifade Mounting Medium and ProLong Diamond Antifade Mounting Medium.

Main properties:

•Prevent dye fading during imaging
•Ready-to-use liquid, can be stored for a long time after mounting
•Mounted samples can Remains stable for months
• Maintains fluorescence signal—virtually no quenching
• Works with Alexa Fluor dye

1.52 refractive index mounting media optimized for imaging with oil immersion objectives Try curing mounting medium Prolong Glass Antifade Mounting Medium or non-curing SlowFade Glass soft anti-fade mounting medium.

ProLong Gold Antifade Mounting Media is available in premixed form for use directly in dropper bottles or 10 mL bottles with or without DNA stain. When DAPI stain binds to DNA, it is excited by ultraviolet light. The excitation wavelength is 360 nm and the maximum emission wavelength is 460 nm. Use DAPI traditional filter for detection. SYTOX Deep Red stain is excited by red light when combined with DNA. The excitation wavelength is 660 nm and the maximum emission wavelength is 682 nm. It is detected using the Cy5/Deep Red traditional filter.

Photobleaching protection without quenching
Many mounting media can destroy up to 80% of the initial fluorescence signal intensity. To create the Molecular Probes Gold Series mounting medium, our scientists developed a chemical additive formula to minimize initial signal quenching of broad-spectrum colors and dyes.

Signal stability during imaging and long-term storage
Our FluoCells range uses epoxy-based mounting media to produce cells that remain stable for years and provide Highest level of anti-fade protection. However, it is inconvenient to apply and requires several steps in organic solvents before sealing. ProLong Gold Antifade Reagent is an excellent alternative for archiving samples. This reagent is also an excellent choice for maintaining signal intensity during prolonged imaging, such as repeated scans under confocal laser scanning illumination conditions. It is easy to use—can be air dried to remove excess moisture and mounting medium. If you want to observe your sample immediately, use our non-curing mounting mediaSlowFade Gold Antifade Mounting Medium.

Excellent index matching without acrylics
Refractive index matching is critical for modern high-resolution imaging applications. ProLong Gold Antifade Mounting Medium has a cured refractive index of 1.47. Additionally, this curing property means you avoid using dirty acrylics (i.e. nail polish), which can damage or destroy modern microscope objectives if they come in contact with them.
