Thermo Scientific 2X RNA Loading Dye is recommended for preparing RiboRuler RNA fractions for agarose or polyacrylamide gel electrophoresis type standards and RNA samples. The solution contains the tracking dyes bromophenol blue, xylene cyanine FF, and the intercalating dye ethidium bromide. In most denaturing agarose gel systems, bromophenol blue migrates slightly faster than human 5S rRNA, while xylene cyanine FF migrates slightly slower than 18S rRNA.

2X RNA loading dye contains the denaturant formamide, which allows RNA fragments to be separated by size even during non-denaturing electrophoresis. Formamide also stabilizes RNA.
