Thermo Scientific 5-fluoroorotic acid (5-fluorouracil-6-carboxylic acid monohydrate; 5-FOA) In the molecular genetics study of yeast, the expression of the URA3 gene (encoding orotate-5'-monophosphate (OMP) dicarboxylase) was detected. Yeast with URA3 gene activity (Ura+) convert 5-FOA into the cytotoxic fluorodeoxyuridine (see figure below). When the medium was supplemented with uracil, only yeast strains with mutations in the URA3 gene could grow in medium containing 5-FOA. 5-FOA is commonly used in experiments with Saccharomyces cerevisiae (URA3), Schizosaccharomyces pombe (URA4 and URA5), Candida albicans (URA3), and Escherichia coli (pyrF).
