The Thermo Scientific λ DNA/HindIII Marker is recommended for sizing large linear double-stranded DNA fragments in agarose gels . λ DNA is completely cleaved with appropriate Thermo Scientific restriction enzymes, purified, and redissolved in storage buffer. DNA fragments contain different sticky ends, the type of which depends on the type of restriction enzyme used to prepare the marker.
The marker can be radioactively labeled using T4 polynucleotide kinase. Additionally, it is available via
Klenow fragment or
Klenow fragment (exo-) Fill-in reaction for radioactive or non-radioactive labeling (
see Resources for protocol).
Features• Determine the size and approximate quantification of macromolecular DNA fragments
• Obvious bands
• Provide matching solutions suitable for sample DNA Loading dye
NoteThe sticky ends of the fragment shown (12 nt cos site of lambda DNA) may anneal and form additional bands. These fragments can be separated by heating at 65°C for 5 minutes, followed by cooling on ice for 3 minutes.
