Ready-to-use Thermo Scientific lambda DNA/HindIII markers are recommended for linear duplex analysis of large molecules in fixed agarose gels. Stranded DNA for size determination. λ DNA is completely cleaved with appropriate Thermo Scientific restriction enzymes, purified, and redissolved in storage buffer. DNA fragments contain different sticky ends, the type of which depends on the type of restriction enzyme used to prepare the marker.

Ready-to-use lambda DNA/EcoRI+HindIII markers are premixed with 6X DNA loading dye for direct loading (after heating) onto agarose gels.

Note
The sticky ends of the identified fragments (12 nt-cos site of lambda DNA) may anneal and form additional bands. These fragments can be separated by heating at 65°C for 5 minutes, followed by cooling on ice for 3 minutes.
