The Phospha-Light™ Assay System provides sensitive detection of secreted alkaline phosphatase (SEAP) reporter proteins in cell culture media. The SEAP gene product is a truncated form of human placental alkaline phosphatase. Secreted reporter proteins do not require the preparation of cell extracts. Cell populations can be monitored over time and kept intact for further experiments.
Patented reagents for ultra-sensitive detection
The Phospha-Light™ detection system is one of the simpler and faster ways to optimize transfection efficiency. The patented reagent combination is the key to the high sensitivity of the Phospha-Light™ test. The system includes CSPD™ high performance alkaline phosphatase substrate, Emerald™ chemiluminescence enhancer and a specific buffer system designed to inhibit non-placental endogenous phosphatase activity. The persistent glow of the chemiluminescence signal can be measured using a photometer without the need for a sample injector.
Application
. The Phospha-Light™ Reporter Assay System has been widely used as a reporter assay to measure gene expression in established cell lines and transfected primary cells, including as a gene knockdown⁄RNA interference readout method. Phospha-Light reporter assays are used for a wide range of viral functional assays, including viral gene expression experiments, viral replication, viral fusogenicity, virus neutralization and virus-mediated cell-cell fusion, and viral infectivity. The use of SEAP reporter proteins is very useful for in vivo detection of reporter genes by detecting the serum of transgenic, transfected, or viral vector-infected animals. The Phospha-Light reporter gene assay system has been used to measure SEAP levels in the serum of transgenic or transfected whole animals, including mouse, rat, marmoset, monkey, and pig serum, and chicken egg allantoic fluid. Recently, mouse SEAP protein (mSEAP) was developed to improve the stability of SEAP in transgenic mice, and the Phospha-Light™ system has been used for sensitive detection of mSEAP.
