Thermo Scientific Pierce TCEP-HCl is a pure, odorless, stable tris(2-carboxyethyl)phosphine crystal. It is a sulfhydryl-free compound that can efficiently reduce the disulfide bonds of proteins and peptides.

Features of TCEP-HCl:

Odorless—Unlike DTT or BME, TCEP has no odor and can easily reduce proteins on the test bench; Contributes to a healthier laboratory environment
Specificity—Selectively and completely reduces the most stable water-soluble disulfides over a wide pH range
• < b>Simple—It can be effectively reduced within five minutes at room temperature and pH 5
Stable—Because of its inherent stability and resistance to air oxidation, there is no need to worry about operation and Take no special precautions during storage; non-volatile and does not react with other functional groups in the protein
Efficient—For most applications, 5 to 50mM TCEP will provide sufficient Molar excess, efficiently reducing disulfide bonds of peptides or proteins within minutes at room temperature
Strong compatibility—Using TCEP, in most applications (such as histidine tagging There is no need to remove the reducing agent before protein purification, maleimide coupling).

Notes on using TCEP:
•TCEP is generally very soluble in aqueous buffers at almost all pHs. Working concentration and 10x stock solutions can therefore be easily prepared in most aqueous buffers.
•TCEP remains stable in aqueous, acidic and alkaline solutions. When TCEP is dissolved directly in water, the obtained pH is approximately 2.5.
•TCEP is not particularly stable in phosphate buffers, especially in neutral or alkaline pH environments. Therefore if using TCEP in PBS buffer, use it immediately after preparation.
•In SDS-PAGE loading buffer, TCEP can replace DTT or 2-mercaptoethanol (2-ME); use TCEP at a final concentration of 50mM.
•Because TCEP is charged in solution, it is not compatible with isoelectric focusing.
