Thermo Scientific EZ-Link Sulfo-NHS-LC-Biotin is a medium length, water-soluble biotinylation reagent. Can be used to label antibodies, proteins and other molecules with primary amines.

EZ-Link Sulfo-NHS-LC-Biotin Features:

Protein Labeling—Biotinylated antibodies facilitate the use of streptoavidin resin or probes for immobilization, purification, or detection
Cell Surface Labeling—Only biotinylate surface proteins of whole cells because negatively charged reagents do not permeate cell membranes
Amine reactivity—Reacts with primary amines (-NH2) such as lysine side chains or amino termini of polypeptides
Soluble—Reacts with common NHS esters The compound's charged sulfo-NHS group improves the water solubility of the reagent
Irreversible—forms a permanent amide bond; the spacer arm is not cleavable
Medium length b>—Spacer arm (total length added to target) is 22.4 Angstroms; provides an excellent balance between sufficient length (minimal steric hindrance for biotin binding) and appropriate mass

Sulfo-NHS- LC-Biotin is one of three very similar EZ-Link reagents that are water-soluble, non-cleavable, and allow simple and efficient biotinization of antibodies, proteins, and any other primary amine-containing macromolecule in solution change. Specific labeling of cell surface proteins is another common application for these unique water-soluble, membrane-impermeable reagents. Different only in spacer arm length, the three Sulfo-NHS ester reagents can optimize labeling and detection experiments in which steric hindrance of biotin binding is an important factor. Sulfo-NHS-LC-Biotin is available in a variety of packaging sizes and as a complete protein labeling kit.

We manufacture biotin reagents to ensure the highest possible level of overall product integrity, consistency and performance for intended research applications.

BiotinN-hydroxysulfosuccinimide (NHS) ester is a widely used biotin chemical reagent type. NHS-activated biotin reacts efficiently with primary amine groups (-NH2) in alkaline buffers to form stable amide bonds. Proteins (such as antibodies) often have several primary amines that can be targeted for labeling, including the side chain of a lysine (K) residue and the N-terminus of each polypeptide.

BiotinThe various NHS ester reagents vary in length, solubility, cell permeability and cleavability. Non-sulfonated NHS-Biotin is cell permeable but must be dissolved in organic solvents such as DMSO or DMF. Sulfo-NHS biotin (and biotin with a PEGylated spacer) can be dissolved directly in water but is not membrane permeable. Various types containing disulfide bonds can be cleaved using reducing agents, thereby separating the biotin groups from the labeled protein.
