Thermo Scientific Pierce DSP (Lomant Reagent) is a water-insoluble, homobifunctional N-hydroxysuccinimide ester (NHS ester) ) cross-linker with thiol cleavability, primary amine reactivity, and is suitable for a variety of applications. DSP has an amine-reactive NHS ester at each end of the 8-carbon spacer arm. NHS esters react with primary amines at pH 7–9 to form stable amide bonds and release N-hydroxysuccinimide. Proteins, including antibodies, often have several primary amines in the side chains of lysine (K) residues and at the N-terminus of each polypeptide, and these primary amines can serve as targets for NHS ester cross-linking reagents. DSP is non-sulfonated and does not dissolve in water, so it must first be dissolved in an organic solvent before being added to the aqueous reaction mixture. Since DSP does not have charged groups, it is lipophilic and membrane permeable, and therefore can be used for intracellular and intramembrane coupling. The sulfonated analog of DSP (DTTSP) is water soluble. If the application requires a stable spacer arm that cannot be cleaved under reducing agent conditions, the non-cleavable analogue of the DSP cross-linker, DSS, can also be used.
