Total Exosome Isolation Reagent (from Plasma) can quickly and efficiently enrich intact exosomes from plasma samples. The kit consists of two parts: total exosome isolation reagent (from plasma) and proteinase K (20 mg/mL). The Total Exosome Isolation Reagent allows simple and reliable concentration of intact exosomes from plasma samples following a protocol that scales with sample volume. Proteinase K is provided separately for optional preliminary plasma processing to remove all plasma proteins, thereby maximizing the purity of the exosome preparation.

Total Exosome Isolation Reagent binds water molecules, thereby forcing poorly soluble components (such as vesicles) out of solution Precipitate, and then these poorly soluble components can be collected by short-term low-speed centrifugation. After 10 min of treatment with proteinase K, the reagent was added to the plasma and the solution was incubated at 2–8°C for 30 min. Precipitated exosomes were recovered by standard centrifugation (10,000 xg, 5 min) at room temperature. The pellet is then resuspended in PBS or similar buffer, and the exosomes are ready for downstream analysis or further purified by affinity methods. Total RNA and proteins are then purified using the Total Exosome RNA and Protein Isolation Kit (Cat. No. 4478545), and intact exosomes can also be used for biological studies involving their pathways, function, and transport.

For downstream mass spectrometry analysis, we recommend gel electrophoresis purification of the isolated exosome suspension prior to analysis.
