Fixable Viability Dye eFluor™ 520 is an irreversible labeling agent prior to cryopreservation, fixation and/or permeabilization procedures Reactive dye for dead cells. Unlike 7-AAD and propidium iodide, cells labeled with Fixable Viability Dye can be washed, fixed, permeabilized, and stained for intracellular antigens without loss of staining intensity from dead cells. Therefore, when studying intracellular targets, use the Fixable Viability Dye to exclude dead cells from the analysis. Fixable Viability Dye can be used to label cells of all species.
Fixable Viability Dye eFluor™ 520 can be excited by the blue laser line (488 nm), has an emission peak of 522 nm, and can be detected using a 530/30 bandpass filter (FITC). Please make sure your instrument can detect the dye. For compensation, it is recommended to use target cell samples stained with Fixable Viability Dye. If the percentage of dead cells is expected to be less than 5%, it is recommended to remove a small aliquot of cells and heat at 65°C for 1 minute, followed immediately by placing on ice for 1 minute. After the above treatment, the heat-inactivated cells can be mixed with viable cells at a ratio of 1:1 and stained using Fixable Viability Dye.
Fixable Viability Dye eFluor™ 520 is supplied as a pre-diluted solution prepared in high quality anhydrous DMSO. Store away from light and moisture. Store with desiccant at temperatures below or equal to -70°C. Can be frozen and thawed up to 20 times. Equilibrate vials to room temperature before opening.
