Accurate

Successful cell cycle analysis requires dyes that are DNA-selective and stain cells in a uniform pattern with minimal fluorescence variability. Vybrant™ DyeCycle™ Purple Stain is ideal for analyzing DNA content in living cells because the stain is cell permeable and, upon binding to double-stranded DNA, emits a fluorescent signal proportional to the DNA mass (see figure).

Low Cytotoxicity

Unlike stains that require high concentrations or chemical structures that are cytotoxic, Vybrant™ DyeCycle™ stains exhibit relatively low Cytotoxicity, sorted according to cell cycle stage.

Minimal compensation

Ideal for commonly used violet laser lines, the maximum fluorescence excitation and emission wavelengths of Vybrant™ DyeCycle™ violet stain/DNA complexes 369/437 nm respectively (see figure).

The violet excitation and narrow emission spectra of Vybrant™ DyeCycle™ Purple Stain make it suitable for multi-pathway analysis as it is compatible with other common dyes (Alexa Fluor™ 488, FITC and RPE) and fluorescent proteins (green fluorescent protein (GFP) and mCherry) have limited spectral overlap. Vybrant™ DyeCycle™ Purple Stain can also be used with UV excitation and emits at ∼440 nm.

Flexible

Vybrant™ DyeCycle™ Purple Stain has been shown to be useful not only on live and fixed cells in cell cycle assays, but also for the identification of mammalian hematopoietic tissue Stem cell side populations and early progenitor cells (shown in the figure).

Simple, robust staining protocol

For cell analysis, simply prepare flow cytometry analysis tubes, each containing 1 mL suspended in complete culture medium cells, the cell concentration is 1 × 106 cells/mL. Add 1 µL of Vybrant™ DyeCycle™ Purple Stain to each tube and mix. The final stain concentration is 5 µM. Incubate for 30 minutes at 37°C in the dark. Keep cells at 37°C until harvest. Samples were analyzed in a flow cytometer using ∼405 nm excitation and ∼440 nm emission wavelengths (without washing or fixation).
