The Molecular Probes™ ATP Assay Kit provides a convenient bioluminescent assay using recombinant firefly luciferase and its substrate D-luciferin to quantitatively determine ATP content. The assay kit is based on the absolute ATP requirements of luciferase for light production (maximum emission wavelength ∼560 nm at pH 7.8). The detection method is extremely sensitive; most chemiluminescence analyzers can detect as low as 0.1 pmol of pre-existing ATP or ATP formed in a kinetic reaction system. This sensitivity enables its use in a wide range of applications, including detection of ATP production in a variety of enzymatic reactions, including ATPase and NADPH oxidase, and detection of low levels in samples such as blood, milk, urine, soil, and sludge. Bacterial contamination. The luciferin–luciferase bioluminescence assay has also been used successfully to study the effects of antibiotics on bacterial populations, to determine cell proliferation and cytotoxicity in bacterial and mammalian cells, and to differentiate the effects of anticancer drugs on malignant cell growth.
