Thermo Scientific Pro-Detect Rapid His Competitive Detection Kit is a single-use, depth-finding lateral flow kit , for quick and easy detection of polyhistidine (His)-tagged proteins in protein expression and purification systems. Lateral flow detection uses membrane-based test strips coated with His tag-specific dye-conjugated antibodies to provide qualitative detection in ten minutes. This assay can be used to detect His-tagged proteins directly from cell culture media (supernatants) or cell lysates without the need for any special instrumentation or handling.

Just add appropriately diluted tissue culture supernatant, lysate, or small purified protein sample to tube or microplate wells and dip lateral flow test strips into each Pro-Detect Rapid Competitive Assay. The gold-coupled detection antibody embedded in the test strip forms a complex with the tagged protein in the sample. In the presence of a target protein tag, the dye-conjugated conjugated antibody will bind to the tagged protein present in the sample at the detection line rather than the protein tag. Therefore, in a positive result, a competitive lateral flow assay will show a decrease in visible test line intensity until all test lines disappear. When the target protein tag is not present, the target-specific dye-conjugated conjugated antibody will bind at the detection line, resulting in a negative result at the visual detection line.
