GeneArt Gibson Assembly EX Master Mix enables DNA assembly using a technology that allows two-step optimized reactions in a single tube Seamlessly join multiple DNA fragments with overlapping sequences. DNA fragments of different lengths can be assembled uniformly through complementary overlapping sequences between fragments. The inherent flexibility and success of this approach is applicable to both small and large DNA vector construction, including the insertion of single and multiple fragments, and has been applied to the construction of entire genomes. The resulting products can be used in a variety of downstream applications, including transformation, PCR, and rolling circle amplification (RCA). The GeneArt Gibson Assembly EX Master Mix Kit includes master mix, positive controls, water, and allows the use of your own competent cells.

GeneArt Gibson Assembly EX Master Mix features include:
Easy—Seamlessly assemble and clone up to 15 DNA fragments in a single reaction
Efficient—Provides reliable and efficient cloning of simple and complex constructs
Flexible—Design guidelines allow assembly into any vector of your choice
Convenient—Master mix kits and cloning kits with chemically or electrically competent cells available
Reliable—over 4,000 scientific publications Citation demonstrates the method's great success

Seamless assembly of up to 15 fragments
GeneArt Gibson Assembly EX cloning is a reliable two-step process in a single tube cloning method that combines up to 15 inserts and vectors in a proprietary enzyme cocktail to assemble DNA fragments with shared end homology without leaving any redundant sequences or scars (seamless ). After the dual optimization reaction is complete, a portion of the assembly reaction is then transformed into chemically or electrocompetently competent cells, resulting in clones that can be used for next-day analysis. The required 20 to 40 base pair fragments with terminal homology can be synthesized de novo by design or can be readily obtained by PCR amplification using custom DNA oligonucleotides. This special enzyme mixture produces seamless and covalently bound high-efficiency DNA constructs.

Provides a reliable method with extremely high efficiency
Because of the covalently bound end product, the GeneArt Gibson Assembly EX method allows the use of chemically or electrocompetent cells to achieve extremely high High transformation efficiency, especially important for challenging constructs. No restriction enzymes, ligation or recombination sites are required, and the method provides a perfectly seamless construct without unwanted redundant bases.

Excellent Versatility
The versatility of the GeneArt Gibson Assembly EX method simplifies many steps by quickly combining, adding, deleting, or exchanging DNA fragments and The subcloning step is eliminated, saving time and effort in the cloning workflow.

Convenient formats
GeneArt Gibson Assembly EX kits are available in a variety of formats, including:
GeneArt Gibson Assembly EX cloning kit, including electrocompetent cells (contains ElectroMAX DH10B electrocompetent cells)
GeneArt Gibson Assembly EX Cloning Kit, Contains Chemically Competent Cells (Contains One Shot TOP10 Chemically Competent Cells)
• GeneArt Gibson Assembly EX Master Mix Kit (cloning kit, without cells) (this page)
