MEGAscript™ T7 Kit is an ultra-high-yield in vitro transcription kit. By changing typical transcription reaction conditions, extremely high nucleotide concentrations can be effectively utilized, resulting in higher yields. T7 Enzyme Mix and 10X Reaction Buffer are specifically calibrated for each batch and RNA polymerase.

Using the MEGAscript™ T7 Kit
The MEGAscript™ T7 Kit contains in vitro transcription reaction components and control templates. The kit will yield a total of 3–5 mg RNA (approximately 100 µg RNA or more/reaction) from the control template provided with the kit. This equates to 400–650 moles of RNA per mole of template. Smaller templates generally produce lower mass and higher molar yield products. New transcription reaction conditions and patented high-yield technology allow RNA synthesis to be 10–50 times greater than that produced by conventional transcription reactions.

Applications
MEGAscript™ T7 Kit is used to synthesize large quantities of unlabeled or low specific activity RNA for a variety of applications, including in vitroTranslation, antisense/microinjection studies, and isolation of RNA-binding proteins. In large-scale transcription reactions, the concentrations of all 4 nucleotides are high, well above the K_m of the enzyme. The RNA yield of the MEGAscript™ T7 Kit is typically more than 10 times higher than that of traditional in vitro transcription reactions. This kit is not recommended for the synthesis of high specific activity probes.

The MessageAmp™ aRNA Amplification Kit is powered by MEGAscript™ technology and is specifically developed to amplify cRNA prior to array analysis. It is recommended to purify in vitro transcripts from free nucleotides, buffer components and enzymes using the MEGAclear™ Kit (Cat. No. AM1908) and the MEGAclear™-96 Kit (Cat. No. AM1909).
