Thermo Scientific DNase I (RNase-free) is an endonuclease that cleaves both single- and double-stranded DNA. It hydrolyzes phosphodiester bonds to produce monodeoxyribonucleotides and oligodeoxyribonucleotides with a 5'-phosphate group and a 3'-OH group.
The activity of this enzyme is strictly dependent on Ca2+ and is activated by Mg2+ or Mn2+ ions.
In the presence of Mg2+, DNase I cleaves each strand of dsDNA independently in a statistically random manner. In the presence of Mn2+, the enzyme cleaves both DNA strands at nearly the same site, producing DNA fragments with blunt ends or overhangs of only one or two nucleotides.
Product Advantages
• Recombinant enzyme
• Purified from non-animal host, low endogenous RNase levels
Applications
• Preparation of DNA-free RNA
• Removal of template DNA after in vitro transcription
• Preparation of DNA-free RNA before RT-PCR and RT-qPCR
• With DNA polymerase I ligation, DNA labeling by nick translation
• DNA-protein interaction studies using DNase I (RNase-free) footprinting
• Generate libraries of randomly overlapping DNA inserts. Use reaction buffer containing Mn2+
Note
DNase I is sensitive to physical denaturation. While mixing, gently invert the tube and shake well. Do not shake violently.
