Thermo Scientific phi29 DNA Polymerase is a high processivity polymerase (greater than 70 kb) with robust Strand displacement activity enables efficient isothermal DNA amplification. phi29 DNA polymerase also has 3'→5 exonuclease (proofreading) activity that preferentially acts on single-stranded DNA or RNA. Therefore, the use of 3'-modified primers is highly recommended.

Product Advantages

• Has the highest synthetic capacity and strand displacement activity among known DNA polymerases – can synthesize DNA fragments exceeding 70kb
•Highly precise DNA synthesis
•Extremely high amplified DNA yields even from minimal amounts of template
•Amplified products can be directly used in downstream applications (PCR, restriction enzyme digestion, SNP genes Typing, etc.)

Applications

• Rolling Circle Amplification (RCA): Generate periodic DNA nanotemplates
• Multiple Displacement Amplification (MDA)
• Genome-wide Unbiased amplification (WGA, see Figure 1 in Supporting Data):
• DNA amplification for SNP and STR detection
• Amplification of cell-free DNA in single cells
• Pathogenic microorganisms or macros Genome
•DNA amplification from filter paper blood spot samples
•DNA template preparation for sequencing
•Protein-primed DNA amplification
•In situ genotyping using padlock probe
br>Recombination-based cloning
• Cell-free cloning of lethal DNA
• RNA-primed DNA amplification

Notes

Add to reaction mixture containing phi29 DNA polymerase Pyrophosphatase may enhance DNA synthesis.
