Thermo Scientific Cfr42I (SacII) restriction enzyme recognizes the CCGC^GG site and cleaves best in B buffer at 37°C (isoschizoases: KspI, SacII, Sfr303I, SgrBI, SstII) . Thermo Scientific Conventional Restriction Endonucleases are a large collection of high quality restriction enzymes optimized to function in one buffer of a five-buffer system. In addition, a universal Tango buffer is provided to facilitate double enzyme digestion. All enzymes demonstrated 100% activity under recommended buffers and reaction conditions. To ensure consistent performance, Thermo Scientific Restriction Enzyme Reaction Buffers contain premixed BSA, which enhances the stability of multiple enzymes and binds contaminants that may be present in DNA preparations.

Note:Cfr42I activity is affected by high salt concentrations. Trace amounts of sodium chloride remaining in the substrate DNA after upstream application may inhibit enzyme activity, resulting in incomplete DNA cleavage. Cfr42I, like its prototype SacII, has difficulty cleaving certain sites in lambda DNA. For efficient cleavage, at least 2 copies of the Cfr42I recognition site are required.
