Thermo Scientific Conventional Restriction Endonucleases are a large collection of high quality restriction enzymes optimized to function in one buffer of a five-buffer system. Universal Tango buffer is also available to facilitate double digestion. All enzymes demonstrated 100% activity under recommended buffers and reaction conditions. To ensure consistent performance, Thermo Scientific restriction enzyme reaction buffers contain premixed BSA, which enhances the stability of multiple enzymes and binds contaminants that may be present in DNA preparations.

Note: Too much enzyme (> 4 U/1 µg DNA) may result in incomplete DNA cleavage. Therefore, we recommend extending the incubation time rather than using too much Bpu10I. Star activity may occur with low salt, high glycerol concentrations (>5%), pH > 7.0, or excess enzyme. Cleavage with Bpu10I requires at least two copies of its recognition sequence.
