Thermo Scientific DpnI restriction endonuclease can recognize the Gm6A^TC site and has better cutting effect in Tango buffer at 37°C (isoschizoase: MalI). Thermo Scientific Conventional Restriction Endonucleases are a large collection of high quality restriction enzymes optimized to function in one buffer of a five-buffer system. In addition, a universal Tango buffer is provided to facilitate double enzyme digestion. All enzymes demonstrated 100% activity under recommended buffers and reaction conditions. To ensure consistent performance, Thermo Scientific Restriction Enzyme Reaction Buffers contain premixed BSA, which enhances the stability of multiple enzymes and binds contaminants that may be present in DNA preparations.

Note:DpnI requires the presence of N6-methyladenine in the recognition sequence to cleave DNA. DNA purified from the dam+ strain will be a substrate for DpnI. DpnI can only cleave completely adenomethylated dam sites. DpnI cleavage at semi-adenomethylated dam sites is 60 times slower. DpnI, Bsp143I, and MboI all recognize the same sequence but have different methylation sensitivities and cleavage sites. Detection was performed using pBR322 DNA.
